Glycobiology of IgE
Gyorgypal A, Banergee S, Conroy M, Anthony R
Immunological Reviews, 2025, vol. 331.1
The importance of glycosylation on IgG function is well established, and the roles of IgE glycans are emerging. This review examines the relationship between IgE glycosylation and its biological function.
Temporal Galactose-Manganese Feeding in Fed-Batch and Perfusion Bioreactors Modulates UDP-Galactose Pools for Enhanced mAb Glycosylation Homogeneity
Gyorgypal A, Fratz-Berilla E, Kohnhorst C, Powers DN, Chundawat SPS
Biotechnology and Bioengineering, 2025
This paper explores how temporal feeding of galactose and manganese in different bioreactor setups can modulate UDP-galactose pools to improve the homogeneity of monoclonal antibody glycosylation.
TAM-ing the beast with IL-34 blockade
Gyorgypal A, Anthony RM
Science Immunology, 2024, vol. 9(101)
FOCIS Highlight: TP53 mutation triggers IL-34 secretion by cancer stem cells, reprogramming macrophages to suppress T cells and promote tumor immune escape.
Evaluation of single-use optical and electrochemical pH sensors in upstream bioprocessing
Fratz-Berilla EJ, Kohnhorst C, Trunfio N, Bush X, Gyorgypal A, Agarabi C
Heliyon, 2024, vol. 10(3)
We found that the single-use optical pH sensors were generally less accurate than the electrochemical probes, especially in detecting large pH drifts from the setpoint.
Automated instant labeling chemistry workflow for real-time monitoring of monoclonal antibody N-glycosylation
Gyorgypal A, Potter OG, Chaturvedi A, Powers DN, Chundawat SP
Reaction Chemistry & Engineering, 2023, vol. 8(10), pp. 2423-2434
We integrated Agilents Instant Procainamide based chemistry workflow into the N-GLYcanyzer PAT unit to allow for nearly 10× faster near real-time analysis of mAb glycoforms.
Recent Advances in Biologic Therapeutic N-Glycan Preparation Techniques and Analytical Methods for Facilitating Biomanufacturing Automation
Tiwold EK, Gyorgypal A, Chundawat SP
Journal of Pharmaceutical Sciences, 2023, vol. 112(6), pp. 1485-1491
we discuss recent advances in mAb sample preparation specifically focused on denaturation, enzymatic processing, and released glycans derivatization methods. Additionally, we review the recent advances in characterization of released and intact N-glycans using chromatography, capillary electrophoresis, and mass spectrometry techniques with a focus on rapid, automated approaches that support analysis of glycosylation profiles of biopharmaceuticals.
Integrated process analytical platform for automated monitoring of monoclonal antibody N-linked glycosylation.
Gyorgypal A, Chundawat SP
Analytical Chemistry, 2022, vol. 94(19), pp. 6986-6995
Here, we developed an online sequential-injection-based PAT system, called N-GLYcanyzer, which can rapidly monitor mAb glycosylation during upstream biomanufacturing.
Recent advances in integrated process analytical techniques, modeling, and control strategies to enable continuous biomanufacturing of monoclonal antibodies
Chopda V, Gyorgypal A, Yang O, Singh R, Ramachandran R, Zhang H, Ierapetritou MG
Journal of Chemical Technology & Biotechnology, 2021
This review article summarizes the recent advances and ongoing obstacles faced during the use of advanced process analytical technologies (PAT), process modeling, and control strategies to enable continuous manufacturing of mAbs. In addition, this review also discusses the process strategies and future directions of advanced continuous manufacturing approaches that have been adapted by other industries and that could be implemented for mAbs production soon.
